Valorização integrada de resíduos queratinosos e subprodutos agroindustriais por basidiomicetos: regulação espécie-dependente da produção enzimática e modulação do perfil bioativo em cultivo submerso

Esta tese teve como objetivo investigar a valorização integrada de resíduos queratinosos e subprodutos agroindustriais por fungos basidiomicetos, enfatizando a regulação espécie-dependente da produção enzimática e a modulação do perfil bioativo da biomassa micelial em cultivo submerso. A proposta se fundamenta no conceito de biorrefinaria fúngica, que busca o aproveitamento integral de resíduos de origem animal e vegetal em processos biotecnológicos de baixo impacto ambiental.No Capítulo I, foram avaliadas dezesseis linhagens de basidiomicetos quanto à capacidade de degradar resíduos queratinosos (pelos e penas) e produzir queratinases e lacases sob diferentes regimes de cultivo (agitado e estático) e fontes de carbono, incluindo melaço de cana-de-açúcar, manitol e glicose. As espécies Lentinus berteroi (U21 2) e Ganoderma stipitatum (U15 4) apresentaram os melhores desempenhos. L. berteroi destacou-se pela maior atividade queratinolítica sob agitação (8,55 U/mL) e atividade específica de 0,18 U/g em meio com melaço, enquanto G. stipitatum exibiu maior eficiência sob condição estática, especialmente em meios contendo pelos. A produção de lacase também variou conforme a espécie e o meio, atingindo até 53,71 U/mL em G. stipitatum cultivado estaticamente com glicose. Esses resultados demonstraram que a produção de enzimas hidrolíticas e oxidativas é fortemente modulada pelo regime de cultivo e pela disponibilidade de oxigênio, refletindo ajustes bioquímicos no secretoma fúngico. O Capítulo II investigou o impacto das condições de cultivo sobre o perfil químico e funcional do micélio de G. stipitatum. O cultivo sob agitação promoveu extratos com maior eficiência antioxidante (IC₅₀ = 2,47 mg/mL para ABTS•⁺), maior fator de proteção solar (FPS = 35,86), manutenção da atividade anticolinesterásica até 0,0078 mg/mL e teores elevados de flavonoides, enquanto o cultivo estático apresentou maior teor global de fenóis. Esses achados indicam reorganização qualitativa do metabolismo secundário dependente da oxigenação. Em conjunto, os resultados comprovam o potencial dos basidiomicetos, especialmente L. berteroi e G. stipitatum, como plataformas eficientes para a conversão sustentável de resíduos agroindustriais em enzimas e metabólitos bioativos de interesse industrial, farmacêutico e cosmético, fortalecendo o papel da biotecnologia fúngica em modelos de bioeconomia circular.

Integrated valorization of keratinous and agro-industrial residues by basidiomycetes: species-dependent regulation of enzymatic production and bioactive profile modulation in submerged culture
This doctoral thesis investigated the integrated valorization of keratinous and agro industrial residues by basidiomycetes, emphasizing species dependent regulation of enzymatic production and modulation of the bioactive profile of mycelial biomass under submerged cultivation. The research is grounded in the concept of a fungal biorefinery, aiming at the full utilization of animal and plant residues through low impact biotechnological processes.In Chapter I, sixteen basidiomycete strains were evaluated for their ability to degrade keratinous residues (hair and feathers) and to produce keratinases and laccases under different cultivation regimes (static and agitated) and carbon sources, including sugarcane molasses, mannitol, and glucose. Lentinus berteroi (U21 2) and Ganoderma stipitatum (U15 4) showed superior performance. L. berteroi exhibited the highest keratinolytic activity under agitation (8.55 U/mL) and a specific activity of 0.18 U/g in molasses medium, while G. stipitatum performed better under static conditions, especially in hair based media. Laccase production also varied according to species and substrate composition, reaching 53.71 U/mL in G. stipitatum cultivated statically with glucose. These findings demonstrate that the production of hydrolytic and oxidative enzymes is strongly modulated by oxygen availability and cultivation regime, reflecting species specific adjustments in fungal secretome composition.Chapter II investigated how cultivation conditions affected the chemical and functional profile of G. stipitatum mycelium. Agitated cultivation produced extracts with higher antioxidant efficiency (IC₅₀ = 2.47 mg/mL in the ABTS radical assay), greater sunscreen potential (SPF = 35.86), and sustained anticholinesterase activity up to 0.0078 mg/mL, while static cultures showed higher total phenolic content. The results indicate a qualitative reorganization of secondary metabolism, driven by oxygenation, leading to the synthesis of more functionally active compounds.Overall, this study demonstrates that cultivation adjustments can strategically direct enzymatic secretion and bioactive metabolite biosynthesis. The integrated approach establishes basidiomycetes—particularly L. berteroi and G. stipitatum—as promising biotechnological platforms for the sustainable conversion of agro industrial residues into high value enzymes and metabolites, reinforcing the role of fungal biotechnology in circular bioeconomy models.
